Phase 3, double-blind trial in 537 adults with type 2 diabetes inadequately controlled on diet and exercise (mean HbA1c 7.9 percent, BMI 35.8, diabetes duration 2.5 years), randomised to once-weekly retatrutide 4, 9 or 12 mg or placebo for 40 weeks at 48 sites in the USA, Mexico and India. HbA1c fell by 1.69, 1.86 and 1.94 percentage points versus 0.81 with placebo (treatment differences -0.88 to -1.12, all p below 0.0001); body weight fell by 11.5, 13.9 and 15.3 percent versus 2.6 percent. Adverse events were mainly mild to moderate gastrointestinal; discontinuation for adverse events 2 to 5 percent; no severe hypoglycaemia.
